The uptake of 5-hydroxytryptamine, histamine, and their amino acid precursors by neoplastic mast cells.

چکیده
Neoplastic mast cells of the mouse not only synthesize histamine and 5-hydroxytryptamine (5-HT)" ' but can take up biogenic amines, 4 a property also demonstrated by normal cells."'6 Previous experiments showed that uptake of 5-HT and histamine,3 but not of the precursor amino acids,2 was inhibited by imipramine, and, moreover, that this drug was more effective in inhibiting uptake of 5-HT than of histamine.' a-Methyldihydroxyphenylalanine inhibited uptake of 5-hydroxytryptophane (5-HTP) but not of histidine2 or of either amine.3 It seemed appropriate to study the effects of other subtances on the uptake of these amines and amino acids in order to see how useful these cells may be for the study of transport. At the same time, it was hoped to gain some insight to factors influencing uptake. Similar studies on amines have been described recently by Day and Stockbridge.'

5-hydroxytryptamine and Histamine as Mediators of the Vascular Injury Produced by Agents Which Damage Mast Cells in Rats
Each of the four agents, ovomucoid, dextran, 48/80, and testis extract, when injected beneath the skin of the dorsa of the paws of rats produces a local vascular injury characterized by a protein-rich edema. Each agent also produces damage to mast cells. Either 5-hydroxytryptamine or histamine produces a response similar in the gross to that elicited by the agents which damage mast cells; howev...
متن کامل5-hydroxytryptamine and Histamine as Mediators of T H E Vascular Injury Produced by Agents Which Damage Mast Cells in Rats* by Donald
No single agent has been found which accounts for the hyperemia and increased vascular permeability associated with many common reactions to injury. Histamine appears to be important in some reactions, but there is abundant evidence that it is not the sole active agent, and in many reactions may not be the major factor. The following experiments demonstrate that the edema produced in the rat by...
متن کاملDeletion of histidine decarboxylase (HDC) enhances the antinociceptive effects of orexin A in the central nervous system
It has long been established that histamine plays a role as a mediator of inflammation. From numerous studies, it has been well known that the amine has many pharmacological actions on a variety of organs. To evaluate the role of histamine in pain perception, we generated HDC knockout mice using a gene targeting method. Histamine is a hydrophilic autacoid, and in most tissues it is stored and s...

متن کاملThe anti-tumor action of lysergic acid derivatives and their serotonin-blocking effect as reflected by the I 131 distribution in rats.
Extensive degranulation and fragmentation of the mesenteric mast cells resulting from a subcutaneous rat sarcoma or its polypeptide have been reported previously (7). Subsequent release of histamine and 5-hydroxytryptamine (serotonin or 5-HT) from these cells interferes with distribution of electrolytes in the body, as illustrated by tracer studies with I m (4), Na ~, and BrS~. 1 Studies showed...
